Orforglipron

Orforglipron is a nonpeptide small-molecule GLP-1 receptor agonist. It belongs to a different chemical class from injectable peptide GLP-1 agonists despite targeting the same receptor family.

What is Orforglipron?

Orforglipron is a nonpeptide small-molecule GLP-1 receptor agonist. It belongs to a different chemical class from injectable peptide GLP-1 agonists despite targeting the same receptor family.

Mechanism & research focus

GLP-1 receptor activation is investigated in glucose regulation and energy-intake pathways. Clinical effects depend on the characterized molecule and formulation, rather than the generic label 'capsulated GLP.'

What the research covers

The 2025 ATTAIN-1 phase 3 trial reported greater weight reduction than placebo over 72 weeks. A separate 2025 phase 3 study in early type 2 diabetes reported greater glycated-hemoglobin reduction. Gastrointestinal adverse events and treatment discontinuations occurred.

What remains uncertain

Weight and glycemic trials answer distinct questions and do not establish every long-term clinical outcome. Published results concern controlled study products. They do not confirm the identity, bioavailability or clinical interchangeability of a separately marketed research capsule.

The evidence label describes the linked research, not the suitability of a catalog product for use in people. Formulation, identity, purity, and measured content must be assessed separately.

Sources & studies
